IGF2BP3 functions as a potential oncogene and is a crucial target of miR-34a in gastric carcinogenesis

Abstract

Background

Gastric cancer (GC) is one of the frequent causes of cancer-related death in eastern Asian population. IGF2BP2 lists in the top rank up-regulated genes in GC, but its functional role is unclear.

Method

The expression of IGF2BP3 in GC cell lines and primary samples was examined by qRT-PCR and Western blot. The biological role of IGF2BP3 was revealed by a series of functional in vitro studies. Its regulation by microRNAs (miRNAs) was predicted by TargetScan and confirmed by luciferase assays and rescue experiments.

Results

IGF2BP3 ranked the No.1 of the up-regulated genes by expression microarray analysis in GC cell lines. The expression level of IGF2BP3 was observed in GC tissues comparing with non-tumorous gastric epitheliums. The up-regulated IGF2BP3 expression was associated with poor disease specific survival. IGF2BP3 knockdown significantly inhibited cell proliferation and invasion. Apart from copy number gain, IGF2BP3 has been confirmed to be negatively regulated by tumor-suppressive miRNA, namely miR-34a. The expression of miR-34a showed negative correlation with IGF2BP3 mRNA expression in primary GC samples and more importantly, re-overexpression of IGF2BP3 rescued the inhibitory effect of miR-34a.

Conclusion

We compressively revealed the oncogenic role of IGF2BP3 in gastric tumorigenesis and confirmed its activation is partly due to the silence of miR-34a. Our findings identified useful prognostic biomarker and provided clinical translational potential.

Emerging roles of lipid metabolism in cancer metastasis

Abstract

Cancer cells frequently display fundamentally altered cellular metabolism, which provides the biochemical foundation and directly contributes to tumorigenicity and malignancy. Rewiring of metabolic programmes, such as aerobic glycolysis and increased glutamine metabolism, are crucial for cancer cells to shed from a primary tumor, overcome the nutrient and energy deficit, and eventually survive and form metastases. However, the role of lipid metabolism that confers the aggressive properties of malignant cancers remains obscure. The present review is focused on key enzymes in lipid metabolism associated with metastatic disease pathogenesis. We also address the function of an important membrane structure-lipid raft in mediating tumor aggressive progression. We enumerate and integrate these recent findings into our current understanding of lipid metabolic reprogramming in cancer metastasis accompanied by new and exciting therapeutic implications.

Successful corticosteroid treatment for pleural effusion due to radiation-induced pleuritis after chemoradiotherapy in a patient with esophageal cancer

Abstract

Concurrent chemoradiotherapy (CRT) is one of the therapies with curative intent used to treat esophageal squamous cell carcinoma (ESCC), and overall survival of patients with esophageal carcinoma treated with CRT has improved. As the number of long-term survivors treated with concurrent CRT has increased, more patients experience late toxicities. A delayed adverse event, such as pleural effusion, is sometimes fatal, but little is known about its treatment. A 72-year-old man diagnosed with ESCC was treated with definitive CRT. He had a complete response to CRT, and 4 years later he complained of dyspnea, caused by pleural effusion. Diuretic agents and drainage of pleural effusion were not sufficiently effective in this case. After oral administration of 30 mg prednisolone, re-accumulation of fluid in the pleural space was controlled and the prednisolone dose was gradually tapered. Corticosteroids could be effective treatment for delayed pleural effusion after radiotherapy, and should be considered an option for treatment-refractory pleural effusion.
